i saw the web and found
http://www.phytotechlab.com/Detail.aspx?ID=1016

Re: TC question

I have no experience in tc.
You can check with Quicklab in Batu Lanchang. I used to buy reagent grade chemical salts there. Mixing MS from component salts is tricky. Probably they have MS. I'll PM you their contact.

Re: TC question

I'll be honest with you guys, I've heard that TC is quite hard to do. Even setting up TC materials can be troublesome and kinda expensive. A few members here do it, but most don't because it requires a depth of knowledge to actually do TC. But it's no harm done if you guys are willing to try. Good luck!

Re: TC question

From what i know, if you really willing to start TC, you will have to bear some problems

1)scolded by your mom or your wife(you need to use pressure cooker-to kill germ, etc)
2)pay lots of electrical bills( you will need artificial light instead of natural life as they are keep in vessel)
3)space(you will have to set up a completely no germ/microorganism condition for tc preparation/separation)
4)distilled water(just as the ingredient)

If you are a student, it is not advisable as it cost a lot and require long patience

Harris Azariel 17th April 2011 07:50 PM

Re: TC question

haha yes that might happen.

But i surely wont be trying it(if i want to try) now since im now very busy with studying for my PMR soon.

Re: TC question

The cheapest startup cost should be a few hundreds. Not too useful for small scale grower. More for learnings I believe.

Bear in mind some chemical substances are hazardous. I wouldn't recommend those haven't been trained in chemistry (till around STPM) to play with it without guidance.

Re: TC question

agreed. Scolding by my mom cant be avoided.....there is a hall that is ideal for a 0bacthria area because there is not much air flow......building a lab wil b my after pmr project.....

Re: TC question

How to do TC ...there are tons of info in the web. print out those you find useful and read them like the holy book:laugh:or jot down the important info.

The mixture - for a beginner to try out, there is no need to use expensive ingredients. Home made medium will do, the same preparation as orchid growers do.what is required, read the following link.

http://www.rv-orchidworks.com/orchid...opagation.html

To sterilise jars(with or without ingredients) and water ordinary pressure cooker is required.

jam jars, beer mugs are excellent. ordinary plastic containers can be used but need to be sterilised before pouring in the cooked ingredients. This has to be done in a sterile chamber ( a laminar flow or glove hood)

different jars that i use
http://i191.photobucket.com/albums/z...u/2f4657fe.jpg

germination in-vitro. nepenthes are difficult and near impossible to TC from tips, stems, roots or leaves. start with seeds first.

http://i191.photobucket.com/albums/z...u/046099d4.jpg

http://i191.photobucket.com/albums/z...u/04be5f5b.jpg

...and i use artificial light placing the jars on window sills, lights are filtered.
http://i191.photobucket.com/albums/z...u/414983cb.jpg

MS and other mixtures, equipments and accessories can be bought from most scientific co.

I use fish aquarium as sterile hood.

Wow Robert, that's very nice and complex! How long did it take for the seeds to germinate?

Harris Azariel 29th April 2011 11:50 PM

Re: TC question

Thanks Robert!

For me now, keeping the media sterile when its out from the microwave is hard since that i cant tight the cap and most of my media contaminated at that part.

before carrying out work the inside hood have to be sterilised using bleach or spirit. The jars with the ingredients are cooked withr pressure cooker for 25 minutes and they are sterile. They are opened only inside the sterilised hood ( do not open outside as that will let contaminants in )

the containers/vessels were bought from a local scientific co.

i made my own medium - sugar, coconut water, charcoal and agar.

I'm in kuching and the co. imported the jars ( and other equipment and accessories ) were imported from, most likely, kuala lumpur.

To a liter coconut water ( or a liter distill water ) i add sugar 20 gm, charcoal 0.5gm, agar 8 gm ( the degree of softnes and watery-like to hardness of the medium depended on the amount of agar, you may play with the amount) 8gm is still soft. Too hard the seedlings roots can't penetrate the medium.

I read and gathered infos from the following link and try it out my own. you may like to prepare your own.
http://www.orchidboard.com/community...ng-medium.html

There is no right or wrong answer. some people add apple juice, banana, papaya, tomato. The ingredients are important to seedlings development else without food the seedlings turn brown and die.

role of charcoal
http://www.ncbi.nlm.nih.gov/pubmed/18786626

for sterilisation i use ethanol, but preferable household bleach(clorox).

is ethanol rubbing alchohol?
hw much does it cost for hw much ethanol?

rubbing alcohol is methanol, contain ethanol but not as pure. yes, can be used to sterilised seeds and the working area ( inner sides of the glove hood and laminar floor)

ethanol was packed in 2.5 lit container.

You need to find the price in KL or anywhere in west malaysia, 'cos we are paying more for most item in the east.

Hi Harris..if you are using the original cap, dont tighten it when they are put into the oven. cap it loosely. After sterilisation wait for them to cool down, tighten the caps as u take them out.
